| 1 Dissolve ingredients except glucose and DTT, then sparge medium with 100% N2 gas for 30 - 45 min to make it anoxic. Adjust pH to 7.2 with HCl, then dispense medium under same gas atmosphere into anoxic Hungate-type tubes or serum vials and autoclave at 121°C for 20 min. Add glucose and DTT from anoxic stock solutions prepared under 100% N2 gas atmosphere and sterilized by filtration. | |||||
