# J13: BL AGAR (GLUCOSE BLOOD LIVER AGAR) ## Main sol. J13 3 g Lab-Lemco beef extract (Oxoid) 10 g Proteose peptone no. 3 (BD-Difco) 5 g Trypticase peptone (BD-BBL) 3 g Phytone peptone (BD-BBL) 5 g Yeast extract (BD-Difco) 150 ml Liver extract (see below) 10 g Glucose 0.5 g Starch (soluble) 10 ml **Solution A** 5 ml **Solution B** 1 g Tween 80 15 g Agar (BD-Difco, Bacto ) 10 ml L-Cysteine HCl x H2O (5%) 50 ml Horse blood 825 ml Distilled water 1. Adjust pH to 7.2. 2. To prepare liver extract, put 10 g liver powder in 170 ml water, keep at 50 to 60C for 1 hr, boil for 5 min, adjust pH to 7.2 and filter. ## Solution A 10 g K2HPO4 10 g KH2PO4 100 ml Distilled water ## Solution B 4 g MgSO4 x 7 H2O 0.2 g NaCl 0.2 g FeSO4 x 7 H2O 0.2 g MnSO4 x n H2O 100 ml Distilled water 1. Mix ingredients except horse blood and adjust pH to 7.2. After autoclaving, cool to 50C, and aseptically add 50.0 ml of horse blood. 2. Comment: Commercial BL agar (Nissui) is also available.
