Boil both solutions under a stream of nitrogen gas for few minutes and autoclave separately under anaerobic conditions at 121C for 15 minutes. Mix aseptically after autoclaving under a stream of sterile nitrogen gas and adjust final pH to 7.8. Distribute in screw-capped bottles or tubes. ## Solution 1 1 g (NH4)2SO4 8.2 g NaCl 0.2 g MgCl2 x 6 H2O 0.05 g CaCl2 x 2 H2O 1 ml **Trace element solution SL-8** 3.6 g Na-DL-malate 1 g Yeast extract 0.5 g Na2S2O3 x 5 H2O 500 ml Distilled water ## Solution 2 500 ml Potassium phosphate buffer (pH 7.8, 20 mM) 1. Boil both solutions under a stream of nitrogen gas for few minutes and autoclave separately under anaerobic conditions at 121°C for 15 minutes. Mix aseptically after autoclaving under a stream of sterile nitrogen gas and adjust final pH to 7.8.
