# 26: ACID RHODOSPIRILLACEAE MEDIUM ## Main sol. 26 0.2 g Yeast extract 1 g Disodium succinate 0.5 g Ammonium acetate 5 ml Fe(III) citrate (0.1% in H2O) 0.5 g KH2PO4 0.4 g MgSO4 x 7 H2O 0.4 g NaCl 0.4 g NH4Cl 0.05 g CaCl2 x 2 H2O 0.4 ml Vitamin B12 (10 mg in 100 ml H2O) 1 ml **Trace element solution SL-6** 0.3 g L-Cysteine HCl 0.5 ml Resazurin (0,1%) 1000 ml Distilled water 1. Adjust pH to 5.7. 2. Boil the medium for a few minute. 3. Sparge medium with 100% N2 gas mixture to make it anoxic. 4. Fill 10 ml in 15 ml tubes with a rubber septum under a stream of nitrogen gas. 5. Autoclave at 121 °C for 20 min. 6. Sterile syringes are used to inoculate and remove samples. 7.
