# 1107: DIALISTER MEDIUM ## Main sol. 1107 10 ml **DIALISTER MEDIUM** 1. To 10 ml of the dialister medium add aseptically: 0.1 ml Na-L-lactate (50% (w/v)) 0.1 ml Haemin (0.5 mg/ml) 0.1 ml NaHCO3 (10% (w/v)) 0.1 ml Glucose (40% (w/v)) 0.05 ml Cystein-HCl x 2 H2O (5% (w/v)) 0.05 ml DTT (100 mM) 0.05 ml **Vitamin K1 solution** 0.5 ml Na-formate (6% (w/v)) 0.5 ml Na-fumarate (6% (w/v)) 0.5 ml Horse blood (sterile) 2. Filter-sterilize all solutions and incubate them in oxygen-free atmosphere. ## DIALISTER MEDIUM 17 g Trypticase (BBL) 3 g Yeast extract 3 g NaCl 3 g KNO3 1000 ml Distilled water 1. Adjust pH to 7.0. 2. Incubate in oxygen-free, 5 - 10% CO2 containing atmosphere. ## Vitamin K1 solution 0.1 ml Vitamin K1 20 ml Ethanol (95 %) 1. Dissolve 0.1 ml of vitamin K1 in 20 ml 95% ethanol and filter sterilize. Store refrigerated in a brown bottle.
