Equipment needed: Autoclave Filter
1 Crucial: prepare all solutions in either clean glassware (incubated in 0.1 % HCl and washed with double distilled water) or GREINER PS container (201172). Do not use container with rubber stoppers or septum.

2 Add 100 mL of 10x FW basal salt solution to 800 mL sterile double distilled water just before use.
3 Add per litre of 1x FW basalt solution: 0.5 mL NH4Cl (1 M), 2 mL NaHCO3 (1 M), 1 mL FeNaEDTA (7.5 mM), 1 mL trace elements solution, 1.95 g MES Hydrat.
4 Adjust pH to the specific pH for each strain.
5 Make up to 1000 ml with double distilled water.
6 Filtrate with 0.2 µm Nalgene® bottle-top sterile filter units (e.g. ThermoFischer 597-4520) in a clean and sterile glass bottle.
7 Aliquote medium in GREINER PS containers. Final medium can be stored at 4 °C in the dark for weeks.

1 Prepare FW basal salt solution (10x) in double distilled water.
2 Autoclave at 121 °C for 20 min.
3 Cool down to room temperature.
4 10x FW basal salt solution can be stored at 4 °C for months.

1 Dissolve 4.2 g of NaHCO3 in 50 mL Double distilled water (use a small beaker with stirring rod to dissolve) and sterilize by filtration (0.2 µm) in the laminar flow hood. Store in a sterile 50 mL falcon tube at 4 °C. Avoid high headspace to liquid ratios to ensure stable concentration.
